{"id":478,"date":"2022-09-22T11:49:55","date_gmt":"2022-09-22T18:49:55","guid":{"rendered":"https:\/\/caam.tech\/?p=478"},"modified":"2022-09-22T11:49:56","modified_gmt":"2022-09-22T18:49:56","slug":"caamtech-creates-two-new-prodrugs-of-4-ho-dipt","status":"publish","type":"post","link":"https:\/\/caam.tech\/caamtech-creates-two-new-prodrugs-of-4-ho-dipt\/","title":{"rendered":"CaaMTech Creates Two New Prodrugs of 4-HO-DiPT"},"content":{"rendered":"\n<p><a href=\"https:\/\/caam.tech\/\">CaaMTech<\/a> scientists, in collaboration with the Manke Lab at UMass Dartmouth, recently reported their synthesis and structural characterization of two novel prodrugs of 4-HO-DiPT.\u00a0 The scientists published their research in a paper titled, \u201c<a rel=\"noreferrer noopener\" href=\"https:\/\/doi.org\/10.1107\/S2056989022009094\" target=\"_blank\">The methanol and ethanol solvates of 4-glutarato-<em>N,N<\/em>-diisopropyltryptamine<\/a>\u201d in the journal <em>Acta Crystallographica<\/em>. These are the first crystal structures reported for any 4-substituted DiPT compound, the first solvates of 4-glutarato-<em>N<\/em>,<em>N<\/em>-diisopropyltryptamine, and the first crystalline material reported for any form of 4-glutarato-<em>N<\/em>,<em>N<\/em>-diisopropyltryptamine.<\/p>\n\n\n\n<p>4-HO-DiPT was first synthesized and characterized as the hydrochloride salt by David Repke in 1977.&nbsp; Its profound psychedelic effects were described by Alexander and Ann Shulgin in their book <em>TiHKAL<\/em>: \u201cI truly doubt that there is another psychedelic drug, anywhere, that can match this one for speed, for intensity, for brevity, and sensitive to dose, at least one that is active orally.&#8221;&nbsp; Shulgin and Shulgin described an onset of about 15 minutes and a duration of action about 2-3 hours.&nbsp; These favorable properties have led to interest in 4-HO-DiPT and its prodrugs as alternatives to psilocybin and other classical psychedelics for use in the clinical treatment of intractable psychiatric conditions.<\/p>\n\n\n\n<p>\u201cFor the tryptamines we\u2019ve studied, the chemical structure is just the beginning of properly identifying the molecule,\u201d explained Dr. Andrew Chadeayne, CaaMTech CEO. \u201cAccurately dosing the compound in biological assays also requires distinguishing between all of the different solvates and crystalline forms because they influence a drug\u2019s molecular weight.\u201d<\/p>\n\n\n\n<p>Though another prodrug of 4-HO-DiPT (4-HO-DiPT glutarate, a \u201chemiester\u201d prodrug of 4-HO-DiPT) was previously reported in <a href=\"https:\/\/patents.google.com\/patent\/US11292765B2\/\">a patent application filed by Field Trip Psychedelics Inc.<\/a> (now Reunion Neuroscience), it was described as an amorphous \u201cwhite cake.\u201d&nbsp; No crystalline material was reported, leaving questions about the identity of the compound in view of widespread ambiguity amidst different crystalline forms of structurally related tryptamines.&nbsp; This latest work reiterates CaaMTech\u2019s commitment to fundamental chemistry and foundational science for the development of psychedelic medicines.<\/p>\n\n\n\n<p>\u201cSynthesizing and characterizing these exciting compounds is an incredible experience, and motivates me to look further into their structures,\u201d said Marilyn Naeem, lead author on the publication. \u201cThe more that we understand their crystalline forms, the more we can understand about their potential as therapeutics.\u201d<\/p>\n","protected":false},"excerpt":{"rendered":"<p class=\"excerpt\">4-HO-DiPT has been proposed as a more clinically-optimized alternative to psilocybin<\/p>\n","protected":false},"author":5,"featured_media":479,"comment_status":"open","ping_status":"closed","sticky":false,"template":"","format":"standard","meta":{"_acf_changed":false,"footnotes":""},"categories":[5],"tags":[],"acf":[],"_links":{"self":[{"href":"https:\/\/caam.tech\/wp-json\/wp\/v2\/posts\/478"}],"collection":[{"href":"https:\/\/caam.tech\/wp-json\/wp\/v2\/posts"}],"about":[{"href":"https:\/\/caam.tech\/wp-json\/wp\/v2\/types\/post"}],"author":[{"embeddable":true,"href":"https:\/\/caam.tech\/wp-json\/wp\/v2\/users\/5"}],"replies":[{"embeddable":true,"href":"https:\/\/caam.tech\/wp-json\/wp\/v2\/comments?post=478"}],"version-history":[{"count":1,"href":"https:\/\/caam.tech\/wp-json\/wp\/v2\/posts\/478\/revisions"}],"predecessor-version":[{"id":480,"href":"https:\/\/caam.tech\/wp-json\/wp\/v2\/posts\/478\/revisions\/480"}],"wp:featuredmedia":[{"embeddable":true,"href":"https:\/\/caam.tech\/wp-json\/wp\/v2\/media\/479"}],"wp:attachment":[{"href":"https:\/\/caam.tech\/wp-json\/wp\/v2\/media?parent=478"}],"wp:term":[{"taxonomy":"category","embeddable":true,"href":"https:\/\/caam.tech\/wp-json\/wp\/v2\/categories?post=478"},{"taxonomy":"post_tag","embeddable":true,"href":"https:\/\/caam.tech\/wp-json\/wp\/v2\/tags?post=478"}],"curies":[{"name":"wp","href":"https:\/\/api.w.org\/{rel}","templated":true}]}}
